1-Cyclopropionyl-d-lysergic acid diethylamide (1cP-LSD) is a lesser-known psychedelic substance belonging to the lysergamide class. It is structurally related to LSD and other analogs such as 1B-LSD, ALD-52, and 1P-LSD. While it is suspected to produce its effects by binding to serotonin receptors in the brain, the precise mechanism remains unidentified.


History and Emergence

The origins of 1cP-LSD are not well-documented. Following the prohibition of 1P-LSD in Germany, 1cP-LSD surfaced on the online research chemical market in 2019. Like its counterparts, it was marketed as a legal alternative to LSD and 1P-LSD.


Subjective Effects

Users of 1cP-LSD report a variety of subjective effects, including open and closed eye visuals, time distortion, conceptual thinking, enhanced introspection, euphoria, and ego loss. A study indicated that when 1cP-LSD is incubated with human serum, it leads to the formation of LSD, suggesting it may function as a prodrug for LSD. Anecdotal evidence supports this theory, with many users experiencing effects that closely resemble those of LSD, albeit with a more gradual onset and smoother cognitive and physical effects.


Chemistry of 1cP-LSD

1cP-LSD is a semisynthetic compound within the lysergamide family. It is named for the cyclopropionyl group attached to the nitrogen of the polycyclic indole structure of LSD. The cyclopropionyl group consists of a carbonyl ring with the chemical formula C3H6 linked to an amino group. The compound features a polycyclic structure that includes a bicyclic hexahydro indole and a bicyclic quinoline group, with an N,N-diethyl carboxamide bound at carbon 8 of the quinoline.


Pharmacological Profile

Due to its structural similarity to LSD, 1cP-LSD is likely a partial agonist at the 5-HT2A receptor, which is thought to mediate its psychedelic effects. Additionally, it may exhibit binding activity at various monoamine receptors, including those for dopamine and norepinephrine, although empirical data to confirm these interactions is currently lacking.


Potential Mechanisms of Action

Given its structural characteristics, it can be hypothesized that 1cP-LSD may not significantly activate dopaminergic pathways, which is typical for most serotonergic psychedelics. Any agonist activity at the D2 receptor could contribute to its psychoactive effects. Furthermore, 1cP-LSD likely binds to most serotonin receptor subtypes, excluding the 5-HT3 and 5-HT4 receptors, although the affinity may be too low for significant activation at typical brain concentrations.


Biased Agonism and Binding Characteristics

1cP-LSD is thought to act as a biased agonist, inducing a receptor conformation that preferentially recruits β-arrestin over activating G proteins. A crystal structure of the 5-HT2B receptor bound to 1cP-LSD reveals an extracellular loop that forms a lid over the diethylamide end of the binding cavity, which may explain the slow unbinding rate from serotonin receptors.
