4-Acetoxy-N-methyl-N-ethyltryptamine, commonly referred to as 4-AcO-MET, Metacetin, or Azomet, is a relatively obscure psychedelic compound belonging to the tryptamine class. This group of substances is known for producing effects similar to those of psilocybin when consumed. Structurally, 4-AcO-MET is related to other psychedelic tryptamines, including 4-AcO-DMT, 4-AcO-DET, and 4-AcO-MiPT.
The chemical structure of 4-AcO-MET is closely aligned with that of 4-HO-MET, leading to the hypothesis that it may function as a prodrug. As a synthetic indole alkaloid, 4-AcO-MET features a core structure typical of tryptamines, which includes a bicyclic indole heterocycle. This structure is connected at the R3 position to an amino group via an ethyl side chain. Notably, 4-AcO-MET is substituted at the R4 position of its indole heterocycle with an acetoxy (AcO) functional group (CH3COO−). Additionally, it possesses a methyl group and an ethyl chain attached to the terminal amine (RN) of its tryptamine backbone.
It has been proposed that 4-AcO-MET is rapidly hydrolyzed into the free phenolic compound 4-HO-MET by serum esterases. However, there is a lack of human studies that investigate the metabolic pathways of this substance. The psychedelic effects of 4-AcO-MET are thought to arise from its action as a partial agonist at the 5-HT2A receptor, although the precise mechanisms behind these interactions and their contribution to the psychedelic experience remain poorly understood.
There is a hypothesis that 4-AcO-MET is quickly converted into 4-HO-MET, which may account for the similarities in their subjective effects. This process is reminiscent of how 4-AcO-DMT is believed to be deacetylated into 4-HO-DMT during the first pass metabolism and subsequent liver processing.
