4-Hydroxy-N-methyl-N-isopropyltryptamine, commonly known as 4-HO-MiPT or Miprocin, is a novel psychedelic compound belonging to the tryptamine class. This substance is known for producing psychedelic effects akin to those of psilocybin when administered. It is part of a broader category of substituted tryptamines, including 4-AcO-DMT, 4-HO-MET, and 4-HO-DiPT, which are recognized for generating variations of the core psychedelic effects associated with psilocin.


Historical Context and Human Use

4-HO-MiPT is relatively uncommon and has a brief history of human consumption. Its psychoactive properties were evaluated by the renowned researcher Alexander Shulgin in 1979, who described a trial involving a 12mg dose as a richly insightful and highly erotic experience. Shulgin documented his findings in his 1997 book, TiHKAL, where he included a description of 4-HO-MiPT. Anecdotal reports and Shulgin's trials suggest that the effects of 4-HO-MiPT are similar to those of psilocin, the active compound found in psilocybin mushrooms.


Pharmacological Properties and Safety

There is limited information regarding the pharmacological properties, metabolism, and toxicity of 4-HO-MiPT in humans. Notably, similar to psilocin, there have been no reported fatalities associated with 4-HO-MiPT use, even in cases where individuals have consumed doses significantly exceeding the active amount. This observation indicates that 4-HO-MiPT is likely well-tolerated from a physiological standpoint.


Synthesis and Chemical Structure

The first synthesis of 4-HO-MiPT was reported in 1981 by a team of chemists led by David Repke. Repke and Shulgin later collaborated on research that examined the effects of various oxygen substituents on the MiPT structure, ultimately describing 4-HO-MiPT as the most intriguing compound in the series, being the only one to exhibit classical hallucinogenic effects. As a synthetic indole alkaloid, 4-HO-MiPT features a core structure typical of tryptamines, characterized by a bicyclic indole heterocycle. It is substituted at the R4 position of its indole heterocycle with a hydroxyl (HO) functional group (OH-), and it contains a methyl group along with an isopropyl chain attached to the terminal amine (RN) of its tryptamine backbone.


Mechanism of Action

The psychedelic effects of 4-HO-MiPT are believed to stem from its action as a partial agonist at the 5-HT2A receptor. However, the precise mechanisms by which these interactions lead to the psychedelic experience remain unclear. It is thought that 4-HO-MiPT functions as a serotonergic psychedelic, with its effects resulting from partial agonism at both the 5-HT2A and 5-HT1A serotonin receptors.
