4-Fluoromethylphenidate (commonly referred to as 4F-MPH) is a novel synthetic stimulant belonging to the phenidate class. It is known for producing long-lasting euphoriant and stimulating effects upon administration. Structurally, it is closely related to the widely prescribed ADHD medication methylphenidate, which is marketed under brand names such as Ritalin and Concerta.
4F-MPH and methylphenidate are believed to share similar pharmacological mechanisms as monoamine reuptake inhibitors. However, anecdotal reports suggest that 4F-MPH exhibits distinctive subjective effects, being considered significantly more euphoric and recreational. Users have reported that it is considerably more potent and associated with fewer uncomfortable side effects, such as anxiety, muscle spasms, and compulsive redosing. This may be attributed to its higher efficiency as a dopamine reuptake inhibitor compared to methylphenidate.
4F-MPH has a relatively short history of recreational use. It was initially developed as a substitute for ethylphenidate, which became illegal in the United Kingdom in April 2015 following a temporary blanket ban.
4F-MPH is a synthetic compound classified under the substituted phenethylamine and substituted phenidate classes, and it is a fluorinated analog of methylphenidate. Like its parent compound, 4F-MPH is a chiral molecule, typically produced as a racemic mixture. Notably, scientific findings suggest that amphetamine analogs containing fluorine, chlorine, bromine, and methyl groups tend to exhibit greater potency than those without.
4F-MPH is believed to primarily function as a dopamine and norepinephrine reuptake inhibitor. This mechanism effectively increases the levels of dopamine and norepinephrine neurotransmitters in the brain by binding to and partially blocking the transporter proteins responsible for clearing these catecholamines from the synaptic cleft. As a result, dopamine and norepinephrine accumulate within the brain's reward pathways, leading to stimulating and euphoric effects. A pharmacological evaluation indicates that the (±)-threo isomer of 4F-MPH is 2.15 times more effective at inhibiting dopamine reuptake and 2.7 times more effective at inhibiting norepinephrine reuptake compared to methylphenidate. Conversely, the (±)-erythro isomer is significantly less effective, being 65 times less effective at dopamine reuptake inhibition and 45.6 times less effective at norepinephrine reuptake inhibition than methylphenidate. Neither racemate of 4F-MPH has a significant impact on serotonin reuptake.
