5-Methoxy-N,N-dimethyltryptamine, commonly known as 5-MeO-DMT, is a naturally occurring psychedelic substance within the tryptamine class. It is structurally related to DMT and 5-HO-DMT (bufotenin) and is reputed to induce some of the most powerful and intense psychedelic states among all known psychedelics. The effects of 5-MeO-DMT are produced through its binding to serotonin receptors in the brain, although the precise mechanism remains unclear.
5-MeO-DMT is found in a variety of plant species as well as in the venom of the toad species Incilius alvarius. While little is known about its historical use, it is believed to have been utilized in South American shamanic practices for centuries. In contemporary settings, both extracts from toad venom and synthetic powder forms are commonly used, primarily through vaporization.
The synthesis and psychoactive effects of 5-MeO-DMT were documented by Alexander Shulgin in his 1997 book, TiHKAL ("Tryptamines I Have Known and Loved"). Recent studies have highlighted its capacity to induce mystical experiences, leading to growing interest in exploring its potential therapeutic effects.
Users report a range of subjective effects from 5-MeO-DMT, including feelings of unity and interconnectedness, time distortion, conceptual thinking, euphoria, and ego loss. The intensity of its psychedelic effects is often compared to DMT, characterized by extreme intensity and very short duration. However, 5-MeO-DMT generally lacks the visual geometry component typical of many psychedelics and tends to produce more physical euphoria and stronger transpersonal or mystical effects, such as ego loss.
Anecdotal reports suggest that 5-MeO-DMT is best suited for individuals with extensive experience using powerful psychedelics, such as DMT, ayahuasca, and DPT, and who are trained in their safe use.
5-MeO-DMT is a ring-substituted indole alkaloid molecule characterized by a core structure typical of tryptamines, which includes a bicyclic indole heterocycle attached at the R3 position to a terminal amine group via an ethyl side chain. It is specifically substituted at the R5 position of its indole heterocycle with a methoxy (MeO) functional group (CH3O−) and contains two methyl groups (CH3-) bound to the terminal amine (RN) of its tryptamine backbone, making it the N-substituted methyl homologue of 5-MeO-MiPT and 5-MeO-DiPT, although it differs significantly in its effects.
The psychedelic effects of 5-MeO-DMT are primarily believed to result from its efficacy as a partial agonist at the 5-HT1A receptor. This compound exhibits a high binding affinity for the 5-HT1A subtype and relatively low binding for the 5-HT2A subtype. Since many common psychedelics are thought to elicit their effects primarily through 5-HT2A receptor activity, the binding profile of 5-MeO-DMT and its contribution to the psychedelic experience remain subjects of ongoing investigation.
Additional mechanisms of action, such as the reuptake inhibition of neurotransmitters like serotonin, noradrenaline, and dopamine, are also thought to play a role. This can lead to dangerous toxicity when 5-MeO-DMT is combined with MAOIs, RIMAs, SSRIs, stimulants, or any substances that act as releasing agents or reuptake inhibitors of monoamine neurotransmitters. Furthermore, 5-MeO-DMT is O-demethylated by the polymorphic cytochrome P450 2D6 (CYP2D6) to an active metabolite, bufotenin.
