6-Allyl-6-nor-lysergic acid diethylamide, also known as N-allyl-nor-lysergic acid N,N-diethylamide, N-allyl-nor-LSD, or simply AL-LAD, is a novel psychedelic substance belonging to the lysergamide class. AL-LAD shares a chemical similarity with LSD and operates primarily by binding to serotonin receptors in the brain.
AL-LAD was first explored in 1984 by researchers Andrew J. Hoffman and David Nichols as part of a series of LSD analogs, which included compounds like ETH-LAD and PRO-LAD. Its effects in humans were later documented by Alexander Shulgin in his book "TiHKAL" ("Tryptamines I Have Known and Loved"), where it was described as "one of the several very potent compounds in a large series of nor-LSD analogues." AL-LAD is a chiral compound with two stereocenters located at R5 and R8. The compound, also referred to as (+)-D-AL-LAD, has an absolute configuration of (5R, 8R). The other three stereoisomers of AL-LAD are not known to possess psychoactive properties. Notably, AL-LAD does not cause a color change with the Marquis, Mecke, or Mandelin reagents, but it does turn purple with Ehrlich's reagent due to the presence of the indole moiety in its structure.
AL-LAD is thought to act as a 5-HT2A partial agonist, with its psychedelic effects likely arising from its efficacy at the 5-HT2A receptors. However, the specific role of these interactions and how they contribute to the psychedelic experience remains an area of scientific inquiry.
AL-LAD shares many characteristics with its parent compound, LSD. In human studies, it appears to be roughly equal in potency, if not slightly less so, while exhibiting a similar mechanism of action.
